Abstract[English]
A method for screening L-nucleic acid aptamers for binding to a target molecule is disclosed. The method comprises: (a) contacting the plurality of L-nucleic acid aptamers with the target molecule under conditions that selectively capture target-bound L-nucleic acid aptamers from the plurality of L- nucleic acid aptamers; (b) amplifying L-nucleic acid aptamers of the target-bound L-nucleic acid aptamers to generate amplified, double-stranded L-nucleic acid oligonucleotides; and (c) isolating amplified double stranded L-nucleic acid oligonucleotides using an electrophoresis based method, thereby screening the plurality of L-nucleic acid aptamers.[French]
L'invention concerne un procédé de criblage d'aptamères d'acide L-nucléique pour identifier la liaison à une molécule cible. Le procédé comprend les étapes consistant à : (a) mettre en contact la pluralité d'aptamères d'acide L-nucléique avec la molécule cible dans des conditions de capture sélective d'aptamères d'acide L-nucléique liés à une cible à partir de la pluralité d'aptamères d'acide L-nucléique ; (b) amplifier des aptamères d'acide L-nucléique des L-aptamères d'acide nucléique liés à une cible pour générer des oligonucléotides d'acide L-nucléique double brin amplifiés ; et (c) isoler des oligonucléotides d'acide L-nucléique double brin amplifiés à l'aide d'un procédé basé sur l'électrophorèse, ce qui permet de cribler la pluralité d'aptamères d'acide L-nucléique.
