Abstract[English] The present invention relates to a method for isolating viable microorganisms which are potentially present in a sample comprising 106 to 108 immune cells or progenitor cells thereof, the method comprising the following steps: - bringing the sample into contact with: • a lysing composition comprising o a lysis buffer comprising a non-ionic detergent at a concentration of between 0.004% and 0.050% of the reaction volume o and/or a lysis solution comprising a saponin at a concentration of between 0.03% and 4% of the reaction volume; • an endonuclease for digesting the nucleic acids released by the action of the lysing composition; • an endopeptidase acting at a pH of between 7 and 8; - separating the viable microorganisms which are potentially present in the sample by filtering the sample through a filter, the pores of which have a diameter of between 0.30 and 0.50 μm. The present invention also relates to the associated solid-phase cytometry detection method.[French] La présente invention concerne un procédé d'isolement de microorganismes viables potentiellement présents dans un échantillon comprenant 106 à 108 cellules immunitaires ou cellules progénitrices de ces dernières, comprenant les étapes suivantes : - Mettre en contact l'échantillon avec • une composition lysante comprenant o un tampon de lyse comprenant un détergent non-ionique à une concentration comprise entre 0,004% et 0,050% du volume réactionnel o et/ou une solution de lyse comprenant une saponine à une concentration comprise entre 0,03% et 4% du volume réactionnel • une endonucléase permettant de digérer les acides nucléiques libérés par l'action de la composition lysante • une endopeptidase agissant à un pH compris entre 7 et 8, - Séparer les microorganismes viables potentiellement présents de l'échantillon par filtration de l'échantillon à travers un filtre dont le diamètre des pores est compris entre 0,30 et 0,50 µm. La présente invention concerne également la méthode associée de détection en cytométrie en phase solide.
