Abstract[English]
Disclosed is a non-invasive method for elevating circulating levels of therapeutically effective mesenchymal stem cells (MSCs). The method comprises (i) selecting, isolating or enriching a sub-population of pluripotent, migratory MSCs co-expressing SSEA3, S1PR2 and CD105 and (ii) administering the enriched cells to the oro-nasopharyngeal tract as a spray, aerosol, liquid, gel, lozenge or patch. The enriched cells exhibit enhanced survival, transendothelial migration and engraftment, overcoming pulmonary sequestration that limits intravenous MSC delivery. Purity tiers of ≥ 10 %, 20 %, 50 %, 75 % and 90 % triple-positive cells allow dose tailoring, and formulations optionally containing cryoprotectants, pro-survival or absorption enhancers. Sources include bone marrow, adipose, or birth tissues. Continuous, intermittent or bolus dosing can be self-administered and is applicable to prophylaxis, systemic regeneration, inflammation modulation and tissue repair. The invention provides a patient-friendly and non-invasive method that safely increases circulating MSC numbers and improves therapeutic efficacy.[French]
L’invention concerne un procédé non invasif d’élévation des niveaux circulants de cellules souches mésenchymateuses (CSM) thérapeutiquement efficaces. Le procédé comprend (i) la sélection, l’isolement ou l’enrichissement d’une sous-population de CSM migrantes multipotentes co-exprimant SSEA3, S1PR2 et CD105 et (ii) l’administration des cellules enrichies au tractus oro-nasopharyngé sous forme de pulvérisation, d’aérosol, de liquide, de gel, de pastille ou de timbre. Les cellules enrichies présentent des caractéristiques améliorées de survie, de migration transendothéliale et de prise de greffe, surmontant la séquestration pulmonaire qui limite l’administration intraveineuse de CSM. Des échelons de pureté ≥ 10 %, 20 %, 50 %, 75 % et 90 % de cellules triples positives permettent un ajustement de la dose, ainsi que des formulations contenant éventuellement des cryoprotecteurs, des promoteurs de survie ou des agents favorisant l’absorption. Les sources comprennent des tissus de moelle osseuse, adipeux ou de naissance. L’administration en continu, intermittente ou en bolus peut être auto-administrée et s’applique à la prophylaxie, à la régénération systémique, à la modulation de l’inflammation et à la réparation tissulaire. L’invention concerne un procédé orienté patient et non invasif, qui augmente en toute sécurité le nombre de CSM circulantes et améliore l’efficacité thérapeutique.
