Abstract[English]
The present disclosure provides an enzyme-free, amplification-free optical method to detect ultra-low concentrations of microbial nucleic acids. The technique relies on detecting high signal-to-noise signals (e.g., fluorescent readouts of hybridization) between target nucleic acid sequences and complementary probes immobilized on the surface of a detection cell. The technique simplifies and accelerates diagnosis workflow, significantly reduces costs, and outperforms existing methods that rely heavily on the use of enzymes, are often time-consuming, and require specialized equipment and trained personnel.[French]
La présente invention concerne un procédé optique sans enzyme et sans amplification permettant de détecter des concentrations ultra-faibles d'acides nucléiques microbiens. La technique repose sur la détection de signaux à haut rapport signal/bruit (par exemple, des signaux fluorescents d'hybridation) entre des séquences d'acides nucléiques cibles et des sondes complémentaires immobilisées à la surface d'une cellule de détection. Cette technique simplifie et accélère le processus de diagnostic, réduit considérablement les coûts et surpasse les procédés existants fortement dépendants de l'utilisation d'enzymes, souvent longs à mettre en œuvre et nécessitant un équipement spécialisé et un personnel qualifié.
