Abstract[English]
The invention provides methods for sequencing RNA molecules from single cells. Initial steps are performed on separate populations of RNA from individual cells, but sequencing and sometimes other steps, such as amplification, are performed after pooling of the populations from individual cells. Sequences can be deconvoluted to their original cells from barcodes added at one or more steps during the process. Single-cell RNA profiles determined by such methods can be useful for characterization of disease, drugs, toxins or environmental materials.[French]
L'invention concerne des procédés de séquençage de molécules d'ARN à partir de cellules uniques. Des étapes initiales sont exécutées sur des populations séparées d'ARN à partir de cellules individuelles, mais le séquençage et parfois d'autres étapes, telles que l'amplification, sont exécutées après le regroupement des populations à partir de cellules individuelles. Des séquences peuvent être déconvoluées vers leurs cellules d'origine à partir de codes à barres ajoutés à une ou plusieurs étapes pendant le processus. Des profils d'ARN unicellulaires déterminés selon de tels procédés peuvent être utiles pour la caractérisation d'une maladie, de médicaments, de toxines ou de matériaux environnementaux.
