Abstract[English]
The present disclosure concerns promoters engineered to increase their responsiveness to a C2 carbon source. The engineered promoters include at least one external carbon source-responsive element (CSRE) located upstream and proximal to the transcription start site of a gene operatively associated thereto. The engineered promoters can be used in a heterologous nucleic acid molecule, a vector, or an expression cassette to promote the expression of a gene in a microbe. The present disclosure also concerns a method for generating the engineered promoters as well as a method for expressing a gene in a recombination microbial host cell using the engineered promoter(s).[French]
La présente invention concerne des promoteurs modifiés pour augmenter leur réactivité à une source de carbone C2. Les promoteurs modifiés comprennent au moins un élément sensible à la source de carbone externe (CSRE) situé en amont et à proximité du site de début de transcription d'un gène associé de manière fonctionnelle à celui-ci. Les promoteurs modifiés peuvent être utilisés dans une molécule d'acide nucléique hétérologue, un vecteur ou une cassette d'expression pour favoriser l'expression d'un gène dans un microbe.
