Abstract[English] In some embodiments, a method for improved sequence coverage of a polypeptide comprises removing interchain disulfide bonds and retaining essentially all intrachain disulfide bonds of a first sample and performing a first mass spectrometry (MS) analysis of the first sample to determine a first amino acid sequence, removing interchain disulfide bonds and intrachain disulfide bonds of a second sample and performing a second MS analysis to determine a second amino acid sequence, and combining the first and second sequence to determine a combined amino acid sequence of the polypeptide.[French] Dans certains modes de réalisation, un procédé de couverture de séquence améliorée d'un polypeptide consiste en l'élimination de ponts disulfure inter-chaînes et la conservation essentiellement de tous les ponts disulfure intra-chaînes d'un premier échantillon, la réalisation d'une première analyse par spectrométrie de masse (MS) du premier échantillon pour déterminer une première séquence d'acides aminés, l'élimination de ponts disulfure inter-chaînes et de ponts disulfure intra-chaînes d'un second échantillon et la réalisation d'une seconde analyse MS pour déterminer une seconde séquence d'acides aminés, et la combinaison des première et seconde séquences pour déterminer une séquence d'acides aminés combinée du polypeptide.
