Abstract[English]
The disclosure provides a population of engineered natural killer cells, wherein the NK cells express at least one polypeptide selected from the group consisting of human leukocyte antigen E, human leukocyte antigen F, and human leukocyte antigen G. The NK cells comprise a genetically engineered disruption of one or more copies of endogenous β-2 microglobulin and a genetically engineered disruption of one or more copies of a human leukocyte antigen class Il-related gene selected from the group consisting of regulatory factor X associated ankyrin containing protein, regulatory factor 5, regulatory factor X associated protein, and class II transactivator. A method of producing human pluripotent stem cell-derived engineered natural killer cells under feeder free conditions also is provided, as is a method of treating a disease or disorder in a subject by administering the NK cells to the subject.[French]
La présente invention concerne une population de cellules tueuses naturelles modifiées, les cellules NK exprimant au moins un polypeptide choisi dans le groupe constitué de l'antigène leucocytaire humain E, de l'antigène leucocytaire humain F et de l'antigène leucocytaire humain G. Les cellules NK comprennent une perturbation par génie génétique d'une ou plusieurs copies de la β-2 microglobuline endogène et une perturbation par génie génétique d'une ou plusieurs copies d'un gène lié à l'antigène leucocytaire humain de classe Il choisi dans le groupe constitué de la protéine contenant de l'ankyrine associée au facteur de régulation X, du facteur de régulation 5, de la protéine associée au facteur de régulation X et du transactivateur de classe II.
