Abstract[English]
Provided herein are systems and methods for the enrichment and detection of nucleic acid molecules. In particular, provided herein are systems and methods for the selective enrichment of a desired nucleic acid molecule in a sample containing non-desired nucleic acid molecules in which a probe with different complementarity to the the desired compared to the undesired nucleic acid, e.g. wild-type and variant. The probe may be bound to the solid phase, e.g. via biotin, the probe binding to the undesired may be cleaved so that the undesired is released and the desired nucleic acid is enriched by binding to the solid phase. The method may also employ differential release from the probe.[French]
L'invention concerne des systèmes et des procédés pour l'enrichissement et la détection de molécules d'acide nucléique. En particulier, l'invention concerne des systèmes et des procédés pour l'enrichissement sélectif d'une molécule d'acide nucléique souhaitée dans un échantillon contenant des molécules d'acide nucléique non souhaitées, une sonde ayant une complémentarité différente avec l'acide nucléique souhaité en comparaison avec l'acide nucléique non souhaité, par exemple de type sauvage et variant. La sonde peut être liée à la phase solide, par exemple, par la biotine, la liaison de la sonde à l'acide nucléique non souhaité peut être clivée de manière que l'acide nucléique non souhaité est libéré et l'acide nucléique souhaité est enrichi par liaison à la phase solide.
