Abstract[English]
Methods and systems that use fluorocarbons for designing scalable living, in-line extraction of heterologous metabolites from microbial cultures are disclosed. The disclosed systems and methods are organism agnostic and be applied to bacteria, yeasts, filamentous microbes, and algae. The disclosed methods and systems use pperfluorocarbon solvent to extract terpenes from engineered microorganisms and allows non-lethal milking of hydrocarbon products from filamentous and single-celled/multi-celled microbial cultures. The disclosed process preferably does not use pressure gradients and relies on the physical interaction of microbes with fluorocarbon at satp and standard temperature of microbial cultivation to partition non-native products. The disclosed process and system also allow for increased production of products providing a physical sink which enables the forward reactions to produce more products than in its absence.[French]
L'invention concerne des procédés et des systèmes utilisant des fluorocarbones pour concevoir une extraction en ligne vivante évolutive de métabolites hétérologues à partir de cultures microbiennes. Les systèmes et les procédés de l'invention sont agnostiques de l'organisme et peuvent être appliqués à des bactéries, des levures, des microbes filamenteux et des algues. Les procédés et systèmes divulgués utilisent un solvant à base de perfluorocarbone pour extraire les terpènes des micro-organismes modifiés et permettent une traite non létale des produits hydrocarbonés des cultures microbiennes filamenteuses et unicellulaires/multicellulaires. Le procédé divulgué n'utilise de préférence pas de gradients de pression et s'appuie sur l'interaction physique des microbes avec le fluorocarbone au satp et à la température standard de la culture microbienne pour séparer les produits non natifs. Le procédé et le système divulgués permettent également une production accrue de produits en fournissant un puits physique qui permet aux réactions en avant de produire plus de produits qu'en son absence.
