Provided herein are skin substitute compositions comprising a stratified epidermis, wherein cells of the stratified epidermis produce, e.g., secrete, a recombinant growth factor and recombinant insulin. In some aspects, the disclosure further relates to methods of manufacturing a skin substitute and methods of using the composition for treatment of a subject, such as for wound healing.[French]
