Abstract[English] The invention relates to cells comprising a shutdown sequence and one or more transgene sequences inserted into both alleles of an intron of an endogenous gene. The invention relates to an optimised process for manufacturing of cells comprising a gene shutdown and, potentially, one or more transgene expression. The invention simplifies the shutdown process and improves shutdown efficiency.[French] L'invention concerne des cellules comprenant une séquence d'inactivation et une ou plusieurs séquences transgéniques insérées dans les deux allèles d'un intron d'un gène endogène. L'invention concerne un procédé optimisé de fabrication de cellules faisant appel à une inactivation génique et, éventuellement, à une ou plusieurs expressions transgéniques. L'invention permet de simplifier le processus d'inactivation et améliore l'efficacité de celle-ci.
