Abstract[English]
The invention relates to the field of predicting (clinical) response of a cancer patient to a therapy comprising an immune checkpoint blocker or to therapy with an immune checkpoint blocker. In particular enriched pre-therapy levels of intra-tumoral CD8 Temra cells and/or intra-tumoral CXCL10-positive macrophages are markers indicative of a positive response. An alternative pre-therapy marker indicative of a positive response relies on detecting enhanced levels of T cell receptor sharing between intra-tumoral and peripheral CD8 Temra cells. The invention therefore refers to methods of e.g. selecting a patient eligible for therapy comprising an immune checkpoint blocker or for therapy with an immune checkpoint blocker including determining the levels of these markers.[French]
L'invention concerne le domaine de la prédiction (clinique) de la réponse d'un patient atteint d'un cancer à une thérapie comprenant un bloqueur de point de contrôle immunitaire ou à une thérapie avec un bloqueur de point de contrôle immunitaire. En particulier, des taux pré-thérapie enrichis de cellules Temra CD8 intratumorales et/ou de macrophages CXCL10-positifs intratumoraux sont des marqueurs indiquant une réponse positive. Un autre marqueur pré-thérapie indiquant une réponse positive repose sur la détection de taux accrus de partage de récepteurs de lymphocytes T entre des cellules Temra CD8 intratumorales et périphériques. L'invention concerne donc des procédés, par exemple, de sélection d'un patient éligible à une thérapie comprenant un bloqueur de point de contrôle immunitaire ou à une thérapie avec un bloqueur de point de contrôle immunitaire comprenant la détermination des taux de ces marqueurs.
