Abstract[English] The present invention relates to the field of glycoproteomics for identifying novel cell surface O-linked glycopeptide epitopes. The invention relates to methods for identifying O-linked glycopeptides, more specifically Tn- or SiaTn-antigen O-glycosylation sites on cell surface proteins, said method comprising a combined workflow of enzymatic treatments of cells or tissue samples to allow specific enrichment and labelling of Tn antigens, followed by mapping of the O-glycopeptides through LC-MS/MS. More specifically, the invention relates to a chemo-enzymatic method to produce labelled O-glycan-peptides from a plasmamembrane-protein extract sample, wherein Sialyltransferase (ST3Gal1/ST3Gal3) treatment is used to protect free galactose against a follow-on treatment with Galactose oxidase, wherein the oxidized glycans are simultaneously labelled. The invention further relates to a tandem mass spectrometry method using an enriched TMT-labelled (Sia)Tn-antigen glycopeptides in a dual fragmentation triggered approach to resolve the identity, localisation and quantification of the O-linked glycan peptides.[French] La présente invention concerne le domaine de la glycoprotéomique en vue de l'identification de nouveaux épitopes glycopeptidiques à liaison O de surface cellulaire. L'invention concerne des procédés d'identification de glycopeptides à liaison O, plus précisément des sites de O-glycosylation d'antigènes Tn- ou SiaTn sur des protéines de la surface cellulaire, ledit procédé comprenant un flux de travail combiné associant des traitements enzymatiques d'échantillons de cellules ou de tissu pour permettre un enrichissement et un marquage spécifiques des antigènes Tn, suivis d'un mappage des O-glycopeptides par LC-MS/MS. Plus précisément, l'invention concerne un procédé chimio-enzymatique de production de O-peptidoglycanes marqués à partir d'un échantillon d'extrait de protéines membranaires plasmatiques, un traitement par la sialyltransférase (ST3Gal1/ST3Gal3) étant utilisé pour protéger le galactose libre contre un traitement de suivi par la galactose oxydase, les glycanes oxydés étant marqués simultanément. L'invention concerne en outre un procédé de spectrométrie de masse en tandem utilisant des glycopeptides d'antigène (Sia)Tn marqués par TMT enrichis dans le cadre d'une démarche déclenchée par une double fragmentation pour déterminer l'identité, la localisation et la quantification des peptidoglycanes à liaison O.
