Abstract[English]
Provided are a method for rapid construction of kidney organoids, and the use thereof. The method comprises: (1) directly inoculating human induced pluripotent stem cells, or aggregating said cells into spheroids and then inoculating same, and culturing the human induced pluripotent stem cells by using a culture medium containing an inhibition component, the inhibition component containing a GSK-3 inhibitor; (2) by means of using an additive-free basic culture medium or a culture medium containing FGF-9, culturing the cells obtained in step (1); (3) by means of using a culture medium containing a GSK-3 inhibitor and FGF-9, culturing the cells obtained in step (2); (4) by means of using a culture medium containing FGF-9, culturing the cells obtained in step (3); and (5) by means of using the additive-free basic culture medium, culturing the cells obtained in step (4). Compared with existing reported differentiation schemes, the present method can shorten the construction time of kidney organoids to 6-8 days, and can be directly applied to in-vitro toxicological tests, thereby achieving an extremely high practical value.[French]
L'invention concerne un procédé de construction rapide d'organoïdes rénaux, ainsi qu'une utilisation associée. Le procédé consiste en : (1) l'inoculation directe de cellules souches pluripotentes induites humaines ou l'agrégation desdites cellules en sphéroïdes, puis l'inoculation de celles-ci, ainsi qu'en la culture des cellules souches pluripotentes induites humaines à l'aide d'un milieu de culture contenant un composant d'inhibition, le composant d'inhibition contenant un inhibiteur de GSK-3 ; (2) la culture, en utilisant un milieu de culture basique sans additif ou un milieu de culture contenant du FGF-9, des cellules obtenues à l'étape (1) ; (3) la culture, au moyen d'un milieu de culture contenant un inhibiteur de GSK-3 et du FGF-9, des cellules obtenues à l'étape (2) ; (4) la culture, au moyen d'un milieu de culture contenant du FGF-9, des cellules obtenues à l'étape (3) ; et (5) la culture, au moyen du milieu de culture basique sans additif, des cellules obtenues à l'étape (4).
