Abstract[English] differentiation of human pluripotent stem cells into definitive endoderm. The mesodermal cell population formation is inhibited by inactivating in the human pluripotent stem cells one or more genes required for the formation of said cell population. The residual human pluripotent stem cells are eliminated or reduced by culturing the human pluripotent stem cells wherein one or more genes required for the formation of the mesodermal cell population has been inactivated in a medium comprising WNT signalling agonist.
