Provided are stationary phase media for adsorption chromatography comprising a plurality of composite microspheres made of a cross-linked polymeric material, each comprising an interior comprising multiple spherical macropores interconnected with one another via interconnecting pores to constitute an open porous network in the interior, and an outer surface comprising a plurality of openings through which the porous network of the interior is in fluid communication with the outer surface and a layer of a hydrophilic polymer covalently attached to the outer surface; methods of making the stationary phase media, and related compositions and methods for chromatographic separation of macromolecules.[French]
L'invention concerne un milieu de phase stationnaire pour chromatographie par adsorption comprenant une pluralité de microsphères composites constituées d'un matériau polymère réticulé, comprenant chacune un intérieur comprenant de multiples macropores sphériques interconnectés les uns aux autres par l'intermédiaire de pores d'interconnexion pour constituer un réseau poreux ouvert à l'intérieur, et une surface externe comprenant une pluralité d'ouvertures à travers lesquelles le réseau poreux de l'intérieur est en communication fluidique avec la surface externe et une couche d'un polymère hydrophile fixée de manière covalente à la surface externe ; l'invention concerne également des procédés de fabrication du milieu de phase stationnaire, et des compositions et des procédés associés pour la séparation chromatographique de macromolécules.
