Abstract[English]
Provided herein are methods for interrogating interactions between a binding molecule and a polypeptide of interest using hydrogen-deuterium exchange mass spectrometry, optionally in combination with in silico modelling of the binding interaction between the binding molecule and the polypeptide. The polypeptides of interest that are interrogated in a disordered domain include, without limitation, androgen receptor polypeptides, alpha-synuclein polypeptides, p27 polypeptides, c-myc polypeptides, and Tau polypeptides. Further described herein are enrichment of molecules that bind to a polypeptide described herein with high affinity, as well as methods of treating diseases or conditions by administering a binding molecule described herein to a subject in need thereof.[French]
L'invention concerne des procédés d'interrogation d'interactions entre une molécule de liaison et un polypeptide d'intérêt à l'aide d'une spectrométrie de masse à échange d'hydrogène-deutérium, éventuellement en combinaison avec une modélisation in silico de l'interaction de liaison entre la molécule de liaison et le polypeptide. Les polypeptides d'intérêt qui sont interrogés dans un domaine désordonné comprennent, sans s'y limiter, des polypeptides de récepteur d'androgène, des polypeptides d'alpha-synucléine, des polypeptides p27, des polypeptides c-myc et des polypeptides Tau.
