Abstract[English]
The present disclosure provides methods of making multiplex targeted gene edits by introducing to DNA molecules or a cell comprising DNA molecules two or more guide RNAs. In some embodiments, the present disclosure provides base editing fusion proteins, protein complexes, compositions, cells and methods of using the same for targeted nucleic acid editing. The base editing fusion protein can comprise a Cas9 domain and a deaminase domain capable of deaminating a nucleotide base in a target nucleic acid sequence.[French]
La présente divulgation concerne des procédés de fabrication d'éditions de gènes ciblés multiplex par introduction, dans des molécules d'ADN ou une cellule comprenant des molécules d'ADN, d'au moins deux ARN guides. Dans des modes de réalisation, la présente divulgation concerne des protéines de fusion d'édition de base, des complexes de protéines, des compositions, des cellules et des procédés d'utilisation associées pour l'édition ciblée d'acides nucléiques.
