Abstract[English] Described herein are methods and compositions for performing genomic editing with multiple gRNAs, also called second-chance CRISPR/Cas9-mediated genome editing (second-chance editing, or scEditing). In certain aspects, the methods described herein comprise use of CRISPR/Cas9 to target genomic sites of interest at least twice sequentially, providing at least one additional opportunity or "second-chance" at each target polynucleotide sequence for the integration of a DNA cargo into by homology-directed repair (HDR) and are useful for increasing the accuracy of editing target polynucleotide sequences by CRISPR-mediated gene editing.[French] La présente invention concerne des procédés et des compositions permettant de réaliser une édition génomique à l'aide de plusieurs ARNg, également appelée édition génomique médiée par CRISPR/Cas9 de seconde chance (édition de seconde chance, ou scEditing). Selon certains aspects, les procédés de la présente invention comprennent l'utilisation de CRISPR/Cas9 pour cibler des sites génomiques d'intérêt au moins deux fois successivement, fournissant au moins une opportunité supplémentaire ou "seconde chance" à chaque séquence de polynucléotide cible pour l'intégration d'une cargaison d'ADN dans celle-ci par la réparation dirigée par homologie (HDR) et sont utiles pour augmenter la précision de l'édition des séquences de polynucléotides cibles par l'édition génique médiée par CRISPR.
