Abstract[English] The invention relates to antigen-binding protein agents specifically binding the hemagglutinin- neuraminidase (HN) protein of human parainfluenza virus 3 (hPIV-3), with cross-reactivity for Sendai Virus (SeV) as well as human parainfluenza virus serotype 1 (hPIV-1) HN, resulting in a cross-neutralizing binding protein to act as a protection agent against parainfluenza infection and/or disease caused by such an infection. More specifically, the invention relates to a family of immunoglobulin single variable domains (ISVDs) capable of broadly blocking HN neuraminidase activity, thereby providing for a novel hPIV-1/-3 cross-neutralizing antiviral protein binding agent. The invention further relates to the use of said antigen-binding proteins in prevention or treatment of acute respiratory tract infections, croup, pneumonia, and exacerbations of chronic obstructive pulmonary disease.[French] L'invention concerne des agents protéiques de liaison à l'antigène se liant de manière spécifique à la protéine hémagglutinine-neuraminidase (HN) du virus parainfluenza humain 3 (hPIV-3), avec une réactivité croisée pour le virus Sendai (SeV) ainsi que le sérotype 1 du virus parainfluenza humain (hPIV-1) HN, conduisant à une protéine de liaison à neutralisation croisée pour agir en tant qu'agent de protection contre une infection parainfluenza et/ou une maladie provoquée par une telle infection. Plus particulièrement, l'invention concerne une famille de domaines variables uniques d'immunoglobuline (ISVD) capables de bloquer largement l'activité de la neuraminidase HN, ce qui permet d'obtenir un nouvel agent de liaison à la protéine antivirale de neutralisation croisée hPIV-1/-3. L'invention concerne en outre l'utilisation desdites protéines de liaison à l'antigène dans la prévention ou le traitement d'infections des voies respiratoires aiguës, du croup, de la pneumonie et des exacerbations de la bronchopneumopathie chronique obstructive.
