Abstract[English]
The present disclosure provides a double-stranded nucleic acid, compositions, and methods relating thereto. The present disclosure encompasses the recognition that structural elements of double stranded nucleic acid, such as stereochemistry of backbone chiral centers (chiral internucleonic linkages), and/or patterns thereof, specially, double-stranded nucleic acid having one or more chirally enriched phosphorothioate internucleotide linkages can have significant impact on oligonucleotide properties and activities, e.g., RNA interference (RNAi) activity, stability, delivery, etc. The present disclosure also provides methods for treatment of diseases using provided double stranded nucleic acid compositions, for example, in RNA interference.[French]
