Abstract[English] Disclosed herein are polypeptides comprising or consisting of anti-Myc single domain antibodies (sdAd), as well as bifunctional and dual-specific bifunctional polypeptides incorporating the anti-Myc sdAbs that induce degradation of a first and optionally a second biological target through the cellular proteasome mechanism. The disclosed bifunctional polypeptides generally comprise a first targeting moiety that includes the anti-Myc sdAb, and a second component that has ubiquitination function, and optionally a second targeting moiety targeting at least a second target protein. The binding of Myc by the first targeting moiety promotes ubiquitination of Myc by the second component, thereby labeling the Myc for proteasomal degradation. These bifunctional polypeptides may be used for the treatment of a disease in a subject, where the first and/or second targeting moieties bind a biological target(s) whose abnormal function and/or expression is associated with the disease.[French] L'invention concerne des polypeptides comprenant ou constitués d'anticorps à domaine unique anti-Myc (sdAd), ainsi que des polypeptides bifonctionnels et bifonctionnels à double spécificité incorporant les sdAbs anti-Myc qui induisent la dégradation d'une première et éventuellement d'une seconde cible biologique à travers le mécanisme de protéasome cellulaire. Les polypeptides bifonctionnels selon l'invention comprennent généralement une première fraction de ciblage qui comprend l'anti-Myc sdAb, et un second composant qui a une fonction d'ubiquitination, et éventuellement une seconde fraction de ciblage ciblant au moins une seconde protéine cible.
