Abstract[English]
The current subject matter provides a cell engineering platform for vector-free and/or viral delivery of payload/cargo compounds and compositions into cells. The platform achieves delivery to cells quickly and in an easy to use manner in an increased environmental temperature. A population of adherent cells is contacted with an aqueous solution including a payload and an alcohol at a concentration (v/v) greater than 2% in an environment having a temperature greater than 25°C. Also provided herein are non-viral methods for reprogramming blood cells to induced pluripotent stem cells (iPSCs). Related apparatus, systems, techniques, articles and compositions are also described.[French]
La présente invention concerne une plate-forme d'ingénierie cellulaire pour l'administration sans vecteur et/ou virale de composés et de compositions de charge utile/cargaison dans les cellules. La plateforme permet d'administrer les cellules rapidement et de manière simple dans un environnement à température élevée. Une population de cellules adhérentes est mise en contact avec une solution aqueuse comprenant une charge utile et un alcool à une concentration (v/v) supérieure à 2 % dans un environnement présentant une température supérieure à 25 °C. La présente invention concerne également des procédés non viraux de reprogrammation de cellules sanguines en cellules souches pluripotentes induites (iPSC).
