Abstract[English]
The present disclosure provides a method for degrading a target protein, comprising: contacting a T-cell with a multispecific binding protein, wherein the multispecific binding protein comprises: a) a first cell surface binding moiety that specifically binds to membrane- bound CTLA-4 on the surface of the T-cell; and b) a second binding moiety that is operatively linked to the first cell surface binding moiety and that specifically binds to the target protein, wherein binding of the multispecific binding protein to the membrane-bound CTLA-4 on the surface of the T-cell facilitates the internalization of the target protein by the T-cell. After internalization, the multispecific binding protein plus the target protein traffic to a lysosome within the T-cell, enabling degradation of the target protein within the lysosome. The present disclosure also provides multispecific binding protein compositions, host cells for making the multispecific binding protein compositions, and methods of using the multispecific binding protein compositions to treat disease.[French]
La présente divulgation concerne un procédé de dégradation d'une protéine cible, comprenant : la mise en contact d'un lymphocyte T avec une protéine de liaison multispécifique, la protéine de liaison multispécifique comprenant : a) une première fraction de liaison de surface cellulaire qui se lie de manière spécifique à CTLA-4 liée à la membrane sur la surface du lymphocyte T ; et b) une seconde fraction de liaison qui est liée de manière fonctionnelle à la première fraction de liaison de surface cellulaire et qui se lie de manière spécifique à la protéine cible, la liaison de la protéine de liaison multispécifique à CTLA-4 liée à la membrane sur la surface de lymphocyte T facilitant l'internalisation de la protéine cible par le lymphocyte T. Après internalisation, la protéine de liaison multispécifique plus le trafic de protéine cible vers un lysosome à l'intérieur du lymphocyte T, permettant la dégradation de la protéine cible à l'intérieur du lysosome. La présente invention concerne également des compositions de protéines de liaison multispécifiques, des cellules hôtes pour fabriquer les compositions de protéines de liaison multispécifiques, et des procédés d'utilisation des compositions de protéines de liaison multispécifiques pour traiter une maladie.
