Abstract[English]
This invention evaluates the anticancer activity of novel bioactive peptides derived from Pistacia species, both in their purified form and when encapsulated in Soy Protein Isolate (SPI) nanoparticles, against various cancer cell lines and non- cancerous cells. Pistacia proteins were enzymatically hydrolyzed, and two new bioactive peptides, Pistacia-Zardin1 (Leu-Ala-Asn-Ala-Arg) and Pistacia-Zardin2 (Val-Asn-Val-Lys-Arg-Ala), were isolated using HPLC. These peptides were encapsulated in SPI nanoparticles via the desolvation method to enhance their stability, solubility, and targeted delivery, achieving average nanoparticle sizes of 100 nm and 110 nm, respectively. Both free and nanoparticle-loaded forms of these peptides demonstrated significant inhibitory activity against OVCAR3, MCF-7, and AGS cancer cell lines without significant toxicity to normal cells, indicating a favorable safety profile compared to traditional chemotherapeutic agents like Doxorubicin. This approach addresses key challenges in cancer treatment by improving the therapeutic potential and delivery of bioactive peptides.[French]
La présente invention porte sur l’évaluation de l'activité anticancéreuse de nouveaux peptides bioactifs dérivés d'espèces de pistacia, à la fois sous leur forme purifiée et lorsqu'ils sont encapsulés dans des nanoparticules d'isolat de protéine de soja (SPI), contre diverses lignées de cellules cancéreuses et des cellules non cancéreuses. Les protéines de pistacia ont été hydrolysées par voie enzymatique, et deux nouveaux peptides bioactifs, Pistacia-Zardin1 (Leu-Ala-Asn-Ala-Arg) et Pistacia-Zardin2 (Val-Asn-Val-Lys-Arg-Ala), ont été isolés par CLHP. Ces peptides ont été encapsulés dans des nanoparticules SPI par l'intermédiaire du procédé de désolvatation pour améliorer leur stabilité, leur solubilité et leur administration ciblée, ce qui permet d'obtenir des tailles moyennes de nanoparticules de 100 nm et 110 nm, respectivement. Les formes libres et chargées de nanoparticules de ces peptides ont démontré une activité inhibitrice significative contre les lignées cellulaires cancéreuses OVCAR3, MCF-7 et AGS sans toxicité significative vis-à-vis des cellules normales, indiquant un profil de sécurité favorable par rapport aux agents chimiothérapeutiques classiques tels que la doxorubicine.
