Abstract[English]
Disclosed herein are methods of screening candidate binding compounds to a plurality of target nucleic acids either of the same sequence or of different sequences utilizing a device configured to detect changes in the folding and unfolding force of the target nucleic acid upon binding to various candidate binding compounds. In the presence of a plurality of nucleic acid sequences, the methods described herein also can rapidly determine the specificity of the small molecule or polypeptide to a nucleic acid structure. Also, disclosed herein are methods of measuring binding kinetics of binding compounds to a target nucleic acid using a device configured to detect changes in the folding and unfolding force of the target nucleic acid upon binding to varying concentrations of the candidate binding compound.[French]
L'invention concerne des procédés de criblage de composés de liaison candidats à une pluralité d'acides nucléiques cibles soit de la même séquence soit de séquences différentes à l'aide d'un dispositif conçu pour détecter les changements dans la force de pliage et de dépliage de l'acide nucléique cible lors de la liaison à divers composés de liaison candidats. En présence d'une pluralité de séquences d'acide nucléique, les procédés décrits ici peuvent également déterminer rapidement la spécificité de la petite molécule ou du polypeptide à une structure d'acide nucléique. L'invention concerne également des procédés de mesure de la cinétique de liaison de composés de liaison à un acide nucléique cible à l'aide d'un dispositif conçu pour détecter des changements de la force de pliage et de dépliage de l'acide nucléique cible lors de la liaison à des concentrations variables du composé de liaison candidat.
