Abstract[English]
Booster immunization compositions and methods of use thereof, are provided. The disclosed compositions use adjuvanted proteins as a booster vaccine targeting functionally relevant pathogen B and T cell epitopes to re-focus a patient's adaptive antibody and cytotoxic T-cell response. The pharmaceutically active ingredient of the re-focusing boost vaccines include of one or several recombinant protein molecules in a full-length or truncated yet functional (meaning the overall antigenic tertiary structure is retained) version of disease-related protein or protein domain, a smaller subunit or specific or modified epitope or combination of shorter epitopes derived from that initial prime antigen sequence. Methods for re-focusing an immune response in a subject, to augment an existing ( not sufficiently protective immune response) and effectively neutralize a pathogen of interest in a mammalian host organism include administering to a subject whose immune system has been primed by a previous infection/ vaccination against the pathogen.[French]
L'invention concerne des compositions d'immunisation de rappel et des méthodes d'utilisation de celles-ci. Les compositions de l'invention utilisent des protéines avec adjuvant en tant que vaccin de rappel ciblant des épitopes de lymphocytes B et T pathogènes fonctionnellement pertinents pour refocaliser un anticorps adaptatif d'un patient et une réponse de lymphocytes T cytotoxiques. L'ingrédient pharmaceutiquement actif des vaccins de rappel de refocalisation comprend une ou plusieurs molécules de protéine de recombinaison dans une version complète ou tronquée mais fonctionnelle (autrement dit la structure tertiaire antigénique globale est conservée) de protéine ou de domaine de protéine liée à une maladie, une sous-unité plus petite ou un épitope spécifique ou modifié ou une combinaison d'épitopes plus courts dérivés de cette séquence d'antigène primaire initiale. L'invention concerne également des méthodes de refocalisation d'une réponse immunitaire chez un sujet, pour augmenter une réponse immunitaire existante (insuffisamment protectrice) et neutraliser de manière efficace un agent pathogène d'intérêt dans un organisme hôte mammifère, comprenant l'administration à un sujet dont le système immunitaire a été activé par une infection/vaccination précédente vis-à-vis du pathogène.
