Abstract[English]
Provided is an engineered ketoreductase polypeptide capable of preparing chiral alcohols, especially for catalyzing the reduction of ethyl 4-chloroacetoacetate to produce ethyl (R) -4-chloro-3-hydroxybutyrate. The polypeptides has high catalytic activity, high stereoselectivity, better thermal stability and solvent tolerance; the reaction process using engineered ketoreductase polypeptides may have a substrate loading of up to 426 g/L and a conversion of up to 99%.[French]
La présente invention concerne un polypeptide de cétoréductase modifié, capable de préparer des alcools chiraux, en particulier de catalyser la réduction du 4-chloroacétate d'éthyle pour produire du (R) -4-chloro-3-hydroxybutyrate d'éthyle. Les polypeptides ont une activité catalytique élevée, une stéréosélectivité élevée, une meilleure stabilité thermique et une meilleure tolérance aux solvants; le processus de réaction utilisant des polypeptides de cétoréductase modifiés peut présenter une charge de substrat allant jusqu'à 426 g/L et un taux de conversion allant jusqu'à 99 %.
