Abstract[English]
Lenalidomide is prescribed for the treatment of patients with malignant tumors and myelodysplastic syndrome. In the present invention, the synthesis technology of key intermediate (S,R)-3-(7-Nitro-3-oxo-1H-isoindole-2-yl)piperidine-2,6-dione and the active pharmaceutical ingredient of lenalidomide was optimized and customized. By carefully evaluating the current synthetic routes, a convenient, practical and cost- effective method for the synthesis of (R,S)-3-(7-nitro-3-oxo-1H-isoindol-2-yl)pyridine- 6,2- Dione as a key intermediate and lenalidomide as an active ingredient were used. At the same time, upon the recrystallization process, the separation and purification steps of the final product were greatly facilitated, and multiple steps of filtration using activated carbon were replaced and filtration device was eliminated, and product waste was reduced in this step. The final washing process of the product obtained according to the optimal method in this invention removed all the impurities and turned the color of the final product into a milky color according to the commercial sample.[French]
Le lénalidomide est prescrit pour traiter des patients atteints de tumeurs malignes et du syndrome myélodysplasique. Dans la présente invention, la technologie de synthèse de l'intermédiaire clé (S,R)-3-(7-nitro-3-oxo-1H-isoindole-2-yl)pipéridine-2,6-dione et du principe actif pharmaceutique lénalidomide a été optimisée et personnalisée. L'évaluation soigneuse des voies de synthèse actuelles permet de mettre en œuvre un procédé commode, pratique et rentable pour la synthèse de (R,S)-3-(7-nitro-3-oxo-1H-isoindol-2-yl) pyridine-6,2-dione en tant qu'intermédiaire clé et de lénalidomide en tant que principe actif. Dans le même temps, lors du processus de recristallisation, les étapes de séparation et de purification du produit final ont été grandement facilitées, et des étapes de filtration multiples utilisant du charbon actif ont été remplacées et le dispositif de filtration a été éliminé. En outre, les déchets de produits ont été réduits dans cette étape. Le procédé de lavage final du produit obtenu selon le procédé optimal de la présente invention élimine toutes les impuretés et fait tourner la couleur du produit final en une couleur laiteuse conformément à l'échantillon commercial.
